Hi, I have been using the mini kit for RNA extraction, and this is one of my early attempts with fungal cells. After using the Precellys, I did a test run with 12 samples and it worked fine — a couple of A260/230 values were a bit off, but generally acceptable for RT-qPCR. I repeated the same steps for a new batch, but now I am getting very low yields (around 20–25 ng/µl), compared to the earlier 600–900 ng/µl. Unless it was divine intervention, nothing has changed from last time. Our kit is close to a year old. Does anyone have suggestions? Could the RNA be stuck in the column, or did nothing bind? So I’ve been trying to find the right ekit for me for a long while now and really love the TD516, but after knowing I’ll need to buy a hi hat stand, kick pedal, and likely a audio interphase if the module sounds alone aren’t good enough, make the combined price is just too much.
